N6-甲基腺嘌呤(N6-methyladenine,m6A)是真核生物中最丰富的RNA化学修饰,在生长发育和疾病发展中扮演重要角色.作为m6A甲基转移酶复合物重要组成部分,甲基化酶样14(methyltransferase like 14,METTL14)通过介导m6A修饰不同分子机制和信号通路中的靶标来参与肿瘤的发生发展过程.本文基于m6A修饰过程,将METTL14在肿瘤发生发展中的作用进行综述,探讨METTL14与肿瘤干细胞增殖更新、肿瘤发生、肿瘤表型转化、肿瘤上皮细胞-间充质转化、自噬、免疫及化学治疗之间的作用机制,并介绍了METTL14在肿瘤中表达和预后的关系,旨在为METTL14在恶性肿瘤的基础研究和临床诊疗提供理论支持.
The roles of methylase like 14 in tumorigenesis and development
N6-methyladenine(m6A)is the most abundant RNA chemical modification in eukaryotes,which plays an important role in growth and disease development.Methyltransferase like 14(METTL14),as an important component of m6A methyltransferase complex,mediates m6A to modify targets in different molecular mechanisms and signaling pathways to participate in the occurrence and development of tumors.Based on the m6A modification process,this paper reviews the role of METTL14 in tumorigenesis and development,and discusses the mechanism of METTL14 and tumor stem cell proliferation and renewal,tumorigenesis,tumor phenotypic transformation,tumor epithelial-mesenchymal transition,autophagy,immunity and chemotherapy.The relationship between the expression of METTL14 in tumors and prognosis was also introduced,aiming to provide theoretical support for the basic research and clinical diagnosis and treatment of METTL14 in malignant tumors.
