Mechanism of transcription factor NRF2 in vascular dementia
Vascular dementia(VD)is a common neurodegenerative disease that is characterized by dementia and a general decline in mental ability.In recent years,the morbidity and mortality of VD have been increasing worldwide,and there are no effective clinical treatment options.A growing number of studies have shown that nuclear factor E2-related factor 2(NRF2)plays an important role in vascular dementia and is involved in regulating the body's antioxidant stress defense system,and its activation enhances the antioxidant capacity of vascular dementia and plays a key role in several aspects of VD pathology.NRF2 is involved in regulating the oxidative stress response to VD injury,reducing neuronal iron death,inhibiting the neuroinflammatory response in microglia,maintaining normal mitochondrial function,and preserving the integrity of the blood-brain barrier.Therefore,this paper reviews the mechanism and research progress of NRF2 in vascular dementia from several aspects,including oxidative stress,neuroinflammation,iron death,mitochondrial damage,and blood-brain barrier,in order to provide new ideas for the treatment of VD.
