脑胶质瘤关键基因表达模式及其预后分析

脑胶质瘤易复发、预后差,但其分子机制尚未得以清晰阐明.本研究基于GEO数据库中的GSE4290和GSE50161数据集,利用GEO2R及微生信等分析工具比较脑胶质瘤与正常脑组织之间的基因表达差异,并对筛选到的差异表达基因进行GO富集分析和KEGG通路富集分析及蛋白互作网络构建.基于蛋白互作网络筛选出排名前10的关键基因,通过LOGpc数据库获得关键基因表达与脑胶质瘤患者的生存关系.Kaplan-Meier分析结果表明,KCNJ9低表达提示脑胶质瘤患者不良预后;GEPIA数据库显示,KCNJ9表达与肿瘤恶性程度相关;通过Timer数据库发现KCNJ9的表达与免疫细胞浸润呈正相关.KCNJ9可以作为脑胶质瘤发展和预后的预测基因,本研究基于生物信息学分析为脑胶质瘤的预后及治疗提供了新的思路.
Expression pattern of key genes in brain glioma and its prognostic analysis
Brain glioma is easy to recur and has poor prognosis.Its molecular mechanism has not been clearly clarified.Based on the GSE4290 and GSE50161 data sets in the GEO database,this study compared the gene expression differences between brain glioma and normal brain tissue using the analysis tools such as GEO2R and bioinformatics,and conducted GO enrichment analysis and KEGG pathway enrichment analysis on the screened differentially expressed genes and the construction of protein interaction network.Based on the protein interaction network,the top 10 key genes were selected,and the relationship between the expression of key genes and the survival of glioma patients was obtained through the LOGpc database.Kaplan Meier analysis results showed that low expression of KCNJ9 suggested poor prognosis in patients with glioma.GEPIA database showed that the expression of KCNJ9 was related to the degree of malignancy of tumor.The expression of KCNJ9 was found to be positively correlated with the infiltration of immune cells through Timer database.KCNJ9 can be used as a predictive gene for the development and prognosis of brain glioma.This study provides a new idea for the prognosis and treatment of brain glioma based on bioinformatics analysis.
