O-连接-N-乙酰氨基葡萄糖化(O-linked β-N-acetylglucosamine,O-GlcNAcylation)是蛋白质翻译后修饰(protein translation modifications,PTMs)的一种,调节各种基本的细胞生物学过程,包括基因转录、信号转导、应激反应和细胞代谢等.目前,O-GlcNAcylation对肿瘤进展的影响己被认可,但其在抗癌疗效中的作用尚未被阐明.因此,本文旨在综述失调的O-GlcNAcylation影响抗癌治疗效果的最新证据,分析O-GlcNAcylation水平失调影响抗癌疗效的几种可能的机制,讨论靶向O-GlcNAcylation作为癌症治疗增敏剂的可能性,为提高抗癌治疗效果提供理论参考.
Implications and mechanisms of O-GlcNAcylation on anti-cancer therapy efficacy
O-linked β-N-acetylglucosamine(O-GlcNAcylation)is one of the protein translation modifications(PTMs)that regulate a variety of fundamental cellular biological processes,including gene transcription,signal transduction,stress response and cell metabolism.The role of dysregulated O-GlcNAcylation in tumorigenesis has been recognized,but its role in the anti-cancer therapy efficacy has not been elucidated.Therefore,this article reviews the latest evidence that dysregulated O-GlcNAcylation affects anti-cancer therapy efficacy,analyzes several possible mechanisms by which dysregulated O-GlcNAcylation levels affect anti-cancer therapy efficacy,and discusses the possibility of targeting O-GlcNAcylation as a cancer therapeutic sensitizer,aiming to provide theoretical references to improve the efficacy of anti-cancer therapy.
