Combined Antitumor Effect of Paclitaxel And Paeonol in the Treatment of Glioma
Objective:To explore and analyze the combined anti-tumor effect of paclitaxel(Ptx)and paeonol(Pae)in the treatment of glioma.Methods:In this study、cell culture、MTT assay、DAPI staining and flow cytometry were used to explore whether the chemotherapy combined with paeonol and paclitaxel can enhance the chemotherapy effect of paclitaxel and reduce the chemotherapy dose of paclitaxel,thus alleviating its side effects.At the same time,the synergistic anti-tumor mechanism of Ptx and Pae was analyzed.Results:The combination of Ptx and Pae inhibited the proliferation of U87 cells in a dose-dependent manner.When Ptx and PAE were combined with dif-ferent concentrations(125μg/ml),the cell survival rate of each concentration group was significantly lower than that of Ptx alone,with statistical significance(P<0.05).The results showed that Pae and Ptx could promote apop-tosis through the mechanism of reactive oxygen species,and Pae and Ptx could promote ROS production and phos-phorylation of protein kinase AKT,and inhibit tumor growth.Conclusion:Pae can further enhance the anti-tumor activity of Ptx against glioma cells,and Pae combined with Ptx can promote apoptosis mainly through the mecha-nism of reactive oxygen species.
