喹乙醇对体外卵母细胞及胚胎发育的影响

喹乙醇(Olaquindox,OLA)作为有效的抗菌药剂,广泛应用于畜禽养殖业。然而,喹乙醇是否对体外卵母细胞及胚胎发育有影响尚不清楚。本实验采用不同浓度喹乙醇(0。1 mM,0。5 mM,1。0 mM,2。0 mM,5。0 mM)对体外获取生发泡(Germinal vesicle,GV)期卵母细胞体外培养,观察卵母细胞的体外成熟率。其次,对实验组体外成熟卵母细胞进行激活发育,统计卵细胞激活率。最后,使用喹乙醇处理体外受精卵观察胚胎发育情况,观察胚胎各阶段发育率并通过荧光定量PCR囊胚期OCT4(Octamer-binding transcription factor 4,OCT4)、CDX2(Caudal type homeobox 2,CDX2)基因表达水平。实验结果显示,1。0 mM、2。0 mM、5。0 mM OLA处理GV期卵母细胞显著阻碍卵母细胞成熟(P<0。05);5。0 mM处理下卵母细胞激活率显著降低(P<0。01)。同时,高剂量的喹乙醇处理下,早期胚胎囊胚发育受阻,相关发育基因OCT4、CDX2 mRNA表达水平呈显著下降。
Effects of olaquindox on oocyte and embryo development in vitro
As an effective antibacterial agent,olaquindox(OLA)has been widely used in livestock and poultry industry.However,it is unclear whether olaquindox has an effect on oocyte and embryo development in vitro.In this study,GV stage oocytes were exposed to different concentrations of olaquindox(0.1,0.5,1.0,2.0,5.0 mM).Observation of oocyte maturation rate after a period of in vitro culture.Secondly,the matured oocytes in vitro were activated and developed,and the activation rate of oocytes was calculated.Finally,the in vitro fertilized eggs were treated with oquindox to observe the embryonic development,the developmental rate of embryos at each stage and the expression levels of OCT4 and CDX2 genes at the blastocyst stage by fluorescent quantitative PCR.The experimental results show that 1.0 mM,2.0 mM,5.0 mM OLA treatment of GV-stage oocytes significantly blocked oocyte maturation(P<0.05).The activation rate of oocytes under 5.0 mM OLA treatment decreased significantly(P<0.01).Meanwhile,the experiment showed that high-dose OLA impaired early embryo development and the mRNA expression levels of related developmental genes OCT4 and CDX2 decreased significantly.
