骆驼蓬在类风湿关节炎大鼠中的抗炎作用及对关节功能、关节滑膜组织NLRP3,ASC,Caspase-1的影响

目的:探讨骆驼蓬在类风湿关节炎大鼠中的抗炎作用及对关节功能、关节滑膜组织NLRP3,ASC,Caspase-1的影响。方法:采用胶原诱导性关节炎模型。将大鼠分为对照组(假手术组)、模型组、骆驼蓬组、扶他林组、甲氨蝶呤组。关节围度及疼痛情况测定;关节滑膜组织HE染色;关节滑膜组织TUNEL染色观察凋亡;免疫组化检测大鼠关节滑膜组织NLRP3,ASC,Caspase-l表达。结果:与空白组相比,模型组、甲氨蝶呤组、骆驼蓬组、扶他林组左右踝关节肿胀程度和关节疼痛指数明显更高(P<0。05);与模型组相比,甲氨蝶呤组、骆驼蓬组、扶他林组左右踝关节肿胀程度和关节疼痛指数明显更低(P<0。05),且骆驼蓬组左右踝关节肿胀程度和关节疼痛指数明显低于甲氨蝶呤组、扶他林组;与空白组相比,模型组、甲氨蝶呤组、骆驼蓬组、扶他林组NLRP3、ASC、Caspase-1明显更高(P<0。05);与模型组相比,甲氨蝶呤组、骆驼蓬组、扶他林组NLRP3、ASC、Caspase-l明显更低(P<0。05),且骆驼蓬组NLRP3、ASC、Caspase-l明显低于甲氨蝶呤组、扶他林组;与空白组相比,模型组、甲氨蝶呤组、骆驼蓬组、扶他林组凋亡率明显更高(P<0。05);与模型组相比,甲氨蝶呤组、骆驼蓬组、扶他林组凋亡率明显更低(P<0。05),且骆驼蓬组凋亡率明显低于甲氨蝶呤组、扶他林组;与模型组比较,甲氨蝶呤组、扶他林组、骆驼蓬组三组关节软骨面均较光滑,滑膜组织增生及炎细胞浸润程度均减轻(P<0。05)。结论:骆驼蓬可通过调节滑膜组织NLRP3炎症小体的表达抑制滑膜组织炎症反应,进而达到改善关节肿胀和疼痛的目的。
Anti-inflammatory Effect of Camel's Fur on Joint Function,Joint Synovial Tissue NLRP3,ASC,Caspase-1 in Rats with Rheumatoid Arthritis
Objective:To investigate the anti-inflammatory effects of camel's bark on joint function,joint synovial tissue NLRP3,ASC,Caspase-1 in rats with rheumatoid arthritis.Methods:Collagen induced arthritis model was adopted.The rats were divided into control group(sham operation group),model group,camelback group,Voltaline group and methotrexate group.Measurement of joint circumference and pain;Joint synovial tissue HE staining;The apoptosis of synovial tissue was observed by TUNEL staining.The ex-pressions of NLRP3,ASC,Caspase-1 in synovial tissues of rats were detected by immunohistochemistry.Results:Compared with blank group,the swelling degree and joint pain index of left and right foot joint in model group,methotrexate group,camelback group and voltaline group were higher(P<0.05).Compared with model group,the left and right foot joint swelling degree and joint pain index in methotrexate group,camphorus group and voltaalin group were lower(P<0.05),and the left and right foot joint swelling degree and joint pain index in camphorus group were significantly lower than those in methotrexate group and Voltaalin group.Compared with blank group,NLRP3,ASC and Caspase-1 in model group,methotrexate group,camelback group and Voltaalin group were higher(P<0.05).Compared with model group,NLRP3,ASC and Caspase-1 were lower in Methotrexate group,Camphorus group and Voltaalin group(P<0.05),and NLRP3,ASC and Caspase-1 in camphorus group were significantly lower than those in methotrexate group and Voltaalin group.Compared with blank group,the apoptosis rate of model group,methotrexate group,camelaria group and voltaline group was higher(P<0.05).Compared with model group,the apoptosis rate of methotrexate group,camel peng group and voltaalin group was signif-icantly lower(P<0.05),and the apoptosis rate of camel peng group was significantly lower than that of Methotrexate group and Voltaalin group.Compared with the model group,the articular cartilage surface of Methotrexate group,Voltaline group and camphor group were smooth,and the degree of synovial tissue hyperplasia and inflammatory cell infiltration were reduced(P<0.05).Conclusion:Camel hair can inhibit the inflammatory response of synovial tissue by regulating the expression of NLRP3 inflammatome,so as to improve joint swelling and pain.
