Analysis of the Relationship between the Expression of LncRNA HOTAIR and miR-17-5p and Proliferation Genes,Clinicopathological Features and Prognosis in Cervical Cancer Tissues
Objective:To investigate the relationship between the expression of long chain non-coding ribonucleic acid(LncRNA)HOX transcriptional antisense RNA(HOTAIR)and microribonucleic acid(miR)-17-5p and proliferation genes,clinicopathological fea-tures and prognosis in cervical cancer tissues.Methods:157 cervical cancer patients who were admitted to The First Affiliated Hospital of Xinjiang Medical University from January 2014 to January 2018 were selected,cervical cancer tissues and cancer adjacent tissues were collected,the expression levels of LncRNA HOTAIR,miR-17-5p and cervical cancer-related proliferation genes PRDX4,NEK2,FHIT and BLCAP were detected by real-time fluorescence quantitative polymerase chain reaction(RT-PCR).The correlation between the ex-pression of LncRNA HOTAIR,miR-17-5p and the expression of proliferation genes in cervical cancer tissues were analyzed by Pearson,the difference in 5-year survival rate of cervical cancer patients with different LncRNA HOTAIR and miR-17-5p expression were ana-lyzed by Kaplan-Meier method.Results:The transcriptional levels of LncRNA HOTAIR,miR-17-5p,PRDX4,NEK2,FHIT and BLCAP in cervical cancer tissues were higher than those in cancer adjacent tissues(P<0.05).The expression of LncRNA HOTAIR and miR-17-5p in cervical cancer tissues was positively correlated with the transcription levels of PRDX4,NEK2,FHIT and BLCAP(P<0.05),the ex-pression of LncRNA HOTAIR was positively correlated with the expression of miR-17-5p(P<0.05).The expression of LncRNA HO-TAIR and miR-17-5p in cancer tissues of International Federation of Gynecology and Obstetrics(FIGO)stage ⅡA,low differentiation,muscular invasion≥ 1/2 and lymph node metastasis was significantly higher than that of FIGO stage I,moderate to high differentiation,muscular invasion<1/2 and no lymph node metastasis(P<0.05).3 cases were lost during the follow-up period,and 132 cases survived.The 5-year overall survival(OS)rate of patients with high expression of LncRNA HOTAIR was lower than that of patients with low ex-pression of LncRNA HOTAIR(P<0.05),and the 5-year OS rate of patients with high expression of miR-17-5p was lower than that of patients with low expression of miR-17-5p(P<0.05).Conclusion:The expression of LncRNA HOTAIR and miR-17-5p in cervical cancer tissues are up-regulate,which are relate to the up-regulation of proliferation gene expression,poor differentiation,high FIGO stage,deep myometrial invasion,lymph node metastasis and low survival rate.
