The Mechanism and Dynamic Change of BDNF/TrkB/CREB Signal of Jinyuhuan Decoction on Mice with Depression and Phlegm-stasis Interformation
Objective:To investigate the effect of Jinyuhuan Decoction on depression and phlegm-stasis in mice and the effects of BDNF/TrkB/CREB signal.Methods:Thirty adult male C57BL/6J mice were selected and divided into blank group,model group,model group,fluoxetine group,low,medium and high dose Jin Yu Tang group.In each group,the mouse model of depression-sputum stasis was established.After the mold production,fluoxetine group was given 3 mg/kg fluoxetine in suspension,low-,medium-and high-dose group was given 0.2 g/kg,0.5 g/kg,0.8 g/kg Jinyu soup,control group and model group were given equal volume of normal saline,1 time/d for 3 weeks.Body mass,sugar water bias,horizontal score,vertical score,experimental motionless time,experimental time,floating time,BDNF/TrkB/CREB mRNA and protein expression levels in the hippocampus.Results:After modeling,the body mass,sugar and water preference,horizontal score,vertical score,BDNF/TrkB/CREB mRNA and protein expression levels in hippocampus of blank group were significantly lower than those of the other 5 groups,and the experimental immovement and floating time were higher than those of the other 5 groups(all P<0.05).After administration,body weight and sugar and water preference in blank group,fluoxetine group,medi-um and high dose Jinyuhuan tang group were higher than those in model group and low dose group,and body weight in low dose group was higher than that in model group(all P<0.05).The horizontal and vertical scores of blank group were higher than those of the other 5 groups,the above indexes of fluoxetine group,medium and high dose Jinyuhuan tang group were higher than those of model group and low dose group,and the low dose group was higher than that of model group(P<0.05).The experimental immobility and floating time of the blank group was significantly lower than that of the other 5 groups,the experimental immobility and floating time of fluoxetine group,medium and high dose Jinyuhuan tang group was lower than that of model group and low dose group,and the low dose group was lower than that of model group(all P<0.05).BDNF/TrkB mRNA level,BDNF,TrkB protein expression level:fluoxetine group>high dose group>medium dose group>low dose group>blank group>model group;There was no difference among fluoxetine group,high dose group and medium dose group(P>0.05),but there was difference among other groups(P<0.05).CREB mRNA level and BDNF protein expression:blank group>fluoxetine group>high dose group>medium dose group>low dose group>model group;There were no dif-ferences among blank group,fluoxetine group,high dose group and medium dose group(P>0.05),but there were differences among other groups(P<0.05).Conclusion:0.5 g/kg Jinyu Huan Decoction can significantly improve the depressive behavior and enhance the repair and regeneration of hippocampus neurons,which may be related to the up-regulation of BDNF/TrkB/CREB signaling pathway in hip-pocampus of mice.
