药物干预PKC-NF-κB通路治疗溃疡性结肠炎的机制研究

目的:研究药物干预PKC-NF-κB通路治疗溃疡性结肠炎的机制。方法:本研究以动物实验研究为主,拟选取33只成年雄性SD大鼠为研究对象,按照研究要求,将SD大鼠分为五组,分别为正常组、低剂量组、中剂量组、高剂量组及UC组,其中,正常组(n=7)、高剂量组(n=6)、中剂量组(n=6)、低剂量组(n=9)、UC组(n=5)。观察五组免疫组化结果、结肠结果、下丘脑结果分析,并进行结肠组织细胞因子相对表达水平,证实药物干预PKC-NF-κB通路治疗溃疡性结肠炎的机制。结果:五组免疫组化检查结果显示,UC组与高剂量组、中剂量组、低剂量组及正常组对比差异有统计学意义(P<0。05),且五组结肠结果及下丘脑检查结果比较差异有统计学意义(P<0。05),同时五组结肠组织细胞因子mRNA相对表达水平对比结果显示,UC组、高剂量组、中剂量组、低剂量组各项指标水平高于正常组(P<0。05),高剂量组各项指标水平高于中剂量组、低剂量组及正常组(P<0。05),而中剂量组高于低剂量组及正常组(P<0。05),低剂量组水平高于正常组(P<0。05)。结论:药物干预PKC-NF-κB通路治疗溃疡性结肠炎效果显著,能够在结肠组织细胞因子mRNA相对表达水平对比分析中也有直接关联,因而可通过药物干预为溃疡性结肠炎治疗提供帮助。
The Mechanism of UC Intervention on PKC-NF-κB Pathway in the Treatment of Ulcerative Colitis
Objective:To investigate the mechanism of UC intervention on PKC-NF-κB pathway in the treatment of ulcerative colitis.Methods:In this study,33 adult male SD rats were selected and divided into five groups according to the research requirements,they were divided into normal group,low-dose group,middle-dose group,high-dose group and UC Group,among which,normal group(N=7),high-dose group(N=6),middle-dose group(N=6),low-dose group(N=9)and UC Group(N=5).The immunohistochemical re-sults,colonic results and hypothalamic results were analyzed.The relative expression levels of cytokines in colon tissue were measured to confirm the mechanism of UC intervention on PKC-NF-κB pathway in the treatment of ulcerative colitis.Results:The results of immuno-histochemistry showed that there were significant differences between UC Group and high-dose group,middle-dose group,low-dose group and normal group(P<0.05),there were significant differences in the results of colonic examination and hypothalamus examination among the five groups(P<0.05),the indexes of UC Group,high dose group,middle dose group and low dose group were higher than those of normal group(P<0.05).The indexes of high dose group were higher than those of middle dose group,low dose group and normal group(P<0.05),the level in the middle dose group was higher than that in the low dose group and the normal group(P<0.05),and the level in the low dose group was higher than that in the normal group(P<0.05).Conclusion:UC intervention of PKC-NF-κB pathway is effective in the treatment of ulcerative colitis,and can be directly related to the relative expression level of cytokine mRNA in colon tis-sue,UC intervention can therefore help in the treatment of ulcerative colitis.
