Study on the Regulatory Effect of High Estrogen Exposure on Placental Damage and Immune Function in Pregnant Rats with Gestational Hypertension
Objective:To investigate the regulatory effect of high estrogen exposure on placental damage and immune function in pregnant rats with gestational hypertension(GH).Methods:GH rat model was constructed,and rats were randomly divided into control group(Control group),GH group,low-dose estrogen group(L-estrogen group),medium-dose estrogen group(M-estrogen group)and high-dose estrogen group(H-estrogen group).The systolic pressure,diastolic pressure,24 h urine protein,follicle stimulating hormone(FSH),estradiol(E2)and luteinizing hormone(LH),tumor necrosis factor-α(TNF-α),interferon-γ(IFN-γ),interleukin(IL)-6,IL-17 contents,and the proportion of helper T cells(Th)17/regulatory T cells(Treg)of pregnant rats were detected.The pathological changes of placental tissue and the ultrastructure of placental trophoblast cells in pregnant rats were observed.Results:The number of villi in GH group was decreased,the nucleus of trophoblast cells was pyknosis,systolic blood pressure,diastolic blood pressure,24 h urine protein,Th17 cell ratio,Th17/Treg ratio,IFN-γ,TNF-α,IL-6 and IL-17 were significantly increased,and the proportion of FSH,E2,LH and Treg cells was significantly decreased(P<0.05).Compared with GH group,the placental tissue damage and nuclear pyknosis of trophoblast cells were reduced in L-estrogen group,M-estrogen group and H-estrogen group,systolic blood pressure,diastolic blood pressure,24 h urine protein,Th17 cell ratio,Th17/Treg ratio,IFN-γ,TNF-α,IL-6 and IL-17 levels were significantly decreased,and the proportion of FSH,E2,LH and Treg cells was significantly increased in a dose-dependent manner(P<0.05).Conclusion:Estrogen exposure can reduce the blood pressure of GH rats,improve placental damage and immune function in GH pregnant rats,and the intervention effect of high-dose estrogen is better.
