血清LncRNA TUG1、miR-29a-3p表达与特发性肺纤维化患者病变程度、肺功能及PI3K/Akt/mTOR信号通路的关系研究

目的:探讨血清长链非编码核糖核酸牛磺酸上调基因1(LncRNA TUG1)、微小核糖核酸-29a-3p(miR-29a-3p)表达与特发性肺纤维化(IPF)患者病变程度、肺功能及磷脂酰肌醇3激酶(PI3K)/蛋白激酶B(Akt)/雷帕霉素靶蛋白(mTOR)信号通路的关系。方法:选取2021年1月至2023年12月期间我院收治的IPF患者52例(研究组),另选取同期于我院体检健康的志愿者50例(对照组)。比较两组血清LncRNA TUG1、miR-29a-3p水平、肺功能参数[肺总量(TLC)、第1秒用力呼气容积占预计值百分比(FEV1%pred)、用力肺活量(FVC)、一氧化碳弥散量占预计值百分比(DLCO%pred)]、高分辨率电子计算机断层扫描(HRCT)评分及PI3K、Akt、mTOR mRNA表达。采用Pearson法分析相关性。结果:研究组血清LncRNA TUG1、PI3K、Akt、mTOR mRNA表达水平高于对照组,miR-29a-3p表达水平低于对照组(P<0。05)。研究组TLC、FEV1%pred、FVC、DLCO%pred低于对照组,HRCT评分高于对照组(P<0。05)。血清 LncRNA TUG1 表达与 HRCT 评分、PI3K、Akt、mTOR mRNA 表达呈正相关,与 TLC、FEV1%pred、FVC、DLCO%pred 呈负相关(P<0。05);miR-29a-3p 与 HRCT 评分、PI3K、Akt、mTOR mRNA 表达呈负相关,与 TLC、FEV1%pred、FVC、DLCO%pred呈正相关(P<0。05)。结论:IPF患者血清LncRNA TUG1呈高表达,miR-29a-3p呈低表达,二者与IPF患者肺组织病变程度、肺功能受损程度及PI3K/Akt/mTOR信号通路异常激活密切相关。
Study on the Relationship between the Expression of Serum LncRNA TUG1 and miR-29a-3p and the Degree of Lesion,Lung Function and PI3K/Akt/mTOR Signaling Pathway in Patients with Idiopathic Pulmonary Fibrosis
Objective:To investigate the relationship between the expression of serum long non-coding RNA taurine up-regulated gene 1(LncRNA TUG1)and microRNA-29a-3p(miR-29a-3p)and the degree of lesion,lung function and phosphatidylinositol 3 kinase(PI3K)/protein kinase B(Akt)/rapamycin target protein(mTOR)signaling pathway in patients with idiopathic pulmonary fibrosis(IPF).Methods:52 IPF patients(study group)admitted to our hospital from January 2021 to December 2023 were selected,and 50 healthy vol-unteers(control group)who underwent physical examination in our hospital during the same period were selected.The levels of serum LncRNA TUG1,miR-29a-3p,lung function parameters[total lung capacity(TLC),forced expiratory volume in the first second as a per-centage of predicted value(FEV1%pred),forced vital capacity(FVC),carbon monoxide diffusion capacity as a percentage of predicted value(DLCO%pred)],high-resolution computed tomography(HRCT)score and PI3K,Akt and mTOR mRNA expression were com-pared in two groups.The correlation was analyzed by Pearson method.Results:The expression levels of serum LncRNA TUG1,PI3K,Akt and mTOR mRNA in study group were higher than those in control group,and the expression level of miR-29a-3p was lower than that in control group(P<0.05).TLC,FEV1%pred,FVC and DLCO%pred in study group were lower than those in control group,and HRCT score was higher than that in control group(P<0.05).The expression of serum LncRNA TUG1 was positively correlated with HRCT score,PI3K,Akt and mTOR mRNA expression,and negatively correlated with TLC,FEV1%pred,FVC and DLCO%pred(P<0.05).And miR-29a-3p was negatively correlated with HRCT score,PI3K,Akt and mTOR mRNA expression,and positively correlated with TLC,FEV1%pred,FVC and DLCO%pred(P<0.05).Conclusion:Serum LncRNA TUG1 is highly expressed,and miR-29a-3p is lowly expressed in IPF patients,the two are closely related to the degree of lung tissue lesions,the degree of lung function damage and the abnormal activation of PI3K/Akt/mTOR signaling pathway in IPF patients.
