Application of Zeolitic Imidazolate Framework-8 Used as a Vaccine Adjuvant/Delivery System
Vaccines are one of the most effective measures to prevent and control disease.At present,the types of vaccines include traditional inactivated vaccines,attenuated vaccines,protein vaccines and nucleic acid vaccines.However,the antigenic components of these vaccines are mostly bioactive substances,which are easily affected by the environment,such as being easily metabolized in the body and having poor immunogenicity,so they need to be used with adjuvants.Now,the traditional adjuvant has some problems such as complicated preparation,low biological safety,poor stability,and low transfection efficiency.Therefore,it is urgent to develop a new vaccine adjuvant/delivery system.ZIF-8(zeolitic imidazolate framework-8),a metal-organic framework,is composed of zinc ions and 2-methylimidazole.It has some advantages including high stability,large specific surface area,low toxicity,and high biocompatibility.Recently,ZIF-8 with the characteristics of unique spatial structure,thermal stability and good biocompatibility has been proved to be a new adjuvant/delivery vector for proteins,nucleic acids,bacteria,and virus vaccines through biomimetic mineralization method.Therefore,the strategies of ZIF-8 encapsulation of different types of vaccines,and the existing problems and prospects were analyzed and summarized in this review,which would provide references for the ZIF-8 as a novel vaccine adjuvant/delivery system.
