Identification and Biological Characterization of a New Broad-spectrum Enterobacter cloacae Bacteriophage ZX14
Enterobacter cloacae is an opportunistic pathogen that can cause serious problems such as sepsis and urinary tract infections in newborns.In this study,we isolated and purified lytic Enterobacter cloacae phage from a fish market in Fujian Province,China.A new lytic Enterobacter cloacae phage ZX14 was isolated and identified.Its biological characteristics,genome analysis,and its application value in the prevention and treatment of multi drug-resistant Enterobacter cloacae infections was explored.The identification results showed that the phage could efficiently lyse Enterobacter cloacae and the titer was as high as 1.5 x 1010 PFU/mL.Analysis of its biological characteristics showed that the incubation period of ZX14 was 20 min and the burst size was 45 PFU/cell when ZX14 was infected with 2025 Enterobacter cloacae strain.It has good stability and adaptability at 50℃,pH 5-12 and other environmental conditions.Genomic analysis showed that the full length of the ZX14 genome was 173 679 bp with 39.92%guanine-cytosine(GC)content.A total of 294 open reading frames(ORFs)and 10 tRNAs were predicted,of which 122 ORFs were structurally similar to known functional proteins.No drug resistance gene and no virulence gene were detected,and the safety was good.In addition,it was found that the phage could also lyse Enterobacter ludwigii and Enterobacter kobei by lytic spectroscopy.In conclusion,this new lytic Enterobacter cloacae phage has a wide range of potential applications and is expected to provide a new solution for the prevention and treatment of Gram Enterobacter cloacae infections.
