烟草青枯菌裂解性噬菌体φPB2的稳定性分析及应用

摘要
为掌握烟草青枯菌噬菌体φPB2的稳定性及其对烟草青枯病的防控效果,测定了温度、pH、盐浓度对其活性的影响,采用离体叶片实验检测了噬菌体对烟草叶片青枯病病斑的防控效果,并通过大田试验检测了田间防控效果.结果表明,噬菌体φPB2在低温和室温水体条件下,30~90 d内比较稳定;在pH为5.0~8.0较稳定;离体叶片接种不同浓度噬菌体液,噬菌体浓度越高,防控效果越好;噬菌体φPB2菌剂田间防控青枯病,青枯病发病率和病情指数较空白对照明显降低,噬菌体菌剂处理平均相对防控率达到70%以上(2021年).研究结果为烟草青枯病生物防控提供一种新策略,为利用烟草青枯菌裂解性噬菌体防控烟草青枯病提供理论依据.
Abstract
In order to study the stability of the lytic bacteriophage φPB2 and its effect against Ralstonia solanacearum,the effects of temperature,pH value,and salt concentration on its activity were measured;the control effect of φPB2 on tobacco bacterial wilt was tested by in vitro leaf experiments,and the control effect was detected by field experiments.The results showed that bacteriophage φPB2 was relatively stable in water at low temperature and room temperature for 1-3 months,and in the pH range of 5.0-8.0.The bacterial wilt disease spots on tobacco leaves after inoculation of different concentrations of the φPB2 showed better prevention and control effects with higher concentration of φPB2 compared to the control;the higher concentration of φPB2,the better effect of prevention and control.Compared with CK,the bacterial wilt rate and disease index of the treatment by the φPB2 were significantly lower,and the average relative control rate reached more than 70%(in 2021).These results may provide a basis for the biological control strategies of bacterial wilt of flue-cured tobacco,and provide a theoretical basis for the use of lytic bacteriophage to control bacterial wilt of tobacco.
