HPLC法测定棕榈酸帕利哌酮原料药中杂质

采用C18 色谱柱(4.6 mm×150 mm,5 μm),流动相A为0.01 mol/L乙酸铵溶液(pH值9.0),流动相B为甲醇-乙腈(体积比 10∶90),梯度洗脱,柱温 25℃,检测波长 280 nm,建立了利用HPLC测定棕榈酸帕利哌酮原料药中杂质的方法.结果显示,主峰与相邻杂质及各已知杂质之间的分离度均大于 1.5,杂质在各自的线性范围内线性关系良好(r>0.9900).本方法专属性强,准确度高,可用于棕榈酸帕利哌酮原料药的质量控制.
Determination of Related Substances of Paliperidone Palmitate Raw Material by HPLC
Objective:To establish a HPLC method for determination of related substances in paliperidone palmitate.Methods:An Inert Sustain C18 column(4.6 mm×150 mm,5 μm)is adopted.The mobile phase A is 0.01 mol/L ammonium acetate solution(pH is adjusted to 9.0)and the mobile phase B is methanol-acetonitrile(the ratio is 10 to 90)with a gradient elution.The column temperature is 25℃and the detection wavelength is set at 280 nm.Results:The separation degree between main peak and adjacent impurities,as well as known impurities,is greater than 1.5;the calibration curves of nine known impurities are linear in the self-concentration range(r>0.9900).Conclusion:The method is specific and accurate,which can be used for quality control of this product.
