摘要

目的:通过对天然高分子的改性,制备多重敏感性的凝胶,以制备可用于药物缓释的载体材料.方法:以大豆分离蛋白(SPI)为原料,添加壳聚糖(CS)与丙烯酰胺(AM),以过硫酸铵(APS)为引发剂,以N,N'-亚甲基双丙烯酰胺(MBA)为交联剂,原位引发聚合,制备SPI/AGC凝胶,并对SPI/AGC水凝胶进行敏感性和体外释放性能测试.结果:SPI/AGC凝胶具备pH敏感性和较好的溶胀性能,溶液pH值越大其溶胀能力越好.对模型药物 5-氟尿嘧啶(5-FU)的负载和体外释放研究表明,在人工肠液中释放 84 h,SPI/AGC对 5-FU的累积释放率达到最大值(67.27%),具有较好的缓释性能.结论:本文制备的SPI/AGC凝胶在生物医药领域具有广泛的应用前景.

Abstract

Objective:To prepare a multi-sensitive gel by modifying natural polymers,and to prepare a carrier material that can be used for drug sustained release.Methods:SPI/AGC gel is prepared by soy protein isolate(SPI)with chitosan(CS)and acrylamide(AM),ammonium persulfate(APS)as initiator,N,N′-methylenebisacrylamide(MBA)as crosslinking agent.The sensitivity and release properties of SPI/AGC hydrogel are tested in vitro.Results:SPI/AGC gel possessed pH sensitivity and better swelling performance.The larger the pH value of solution,the better the swelling ability.The load and in vitro release of the model drug 5-fluorouracil(5-FU)show that the cumulative release rate of 5-FU reache the maximum value(67.27%)by SPI/AGC after 84 h release in artificial intestinal fluid,which shows good sustained release performance.Conclusion:SPI/AGC gel prepared in this paper has a wide application prospect in the field of biomedicine.
