腰痹通胶囊中主要成分在不同种属肝微粒体中代谢物谱及稳定性研究

Research on Differences of Metabolite Profiling and Metabolic Stability of the Main Ingredients of Yao-Bi-Tong Capsule in the Different Species of liver Microsomes

本研究旨在应用肝微粒体孵育技术研究腰痹通胶囊中主要成分在不同种属肝微粒体(人、食蟹猴、比格犬、大鼠和小鼠)中I相代谢物谱及稳定性差异.采用UPLC-Q-TOF技术寻找腰痹通胶囊孵育样品中可能的代谢产物,采用UPLC-TQ技术测定腰痹通胶囊孵育样品中主要成分的含量变化.结果表明,腰痹通胶囊中蛇床子素在不同种属肝微粒中的I相代谢物谱和代谢稳定性均有明显差异,而其他主要成分在不同种属肝微粒体中代谢较稳定,代谢差异不明显.
This study aimed to investigate phase I metabolism of the main ingredients of Yao-Bi-Tong Capsule and compare the difference of their metablite profile and metablic stability in the different species of liver microsomes(human,cynomolgus monkeys,beagle dogs,SD rats and mouse)through liver microsomal incubation technique.UPLC-Q-TOF technology is used to find the possible metabolites in the incubated samples.UPLC-TQ technology is used to determine the change of the content of the main components in the incubated samples.The results show that phase I of osthole has some significant differences on the metablite profile and metabolic stability in five different species of liver microsomes.However,metabolism of other main ingredients is relatively stable and their metabolic difference are not significant in different species of liver microsomes.
