Objective:To investigate the effect of polysaccharide of Polygonatum odoratum in culture medium on human umbilical cord mesenchymal stem cells(hUC-MSCs)culture.Methods:Compare the morphological,stability and yield of human umbilical cord mesenchymal stem cells obtained by adding polysaccharide of Polygonatum odoratum in culture medium and traditional culture method.Results:The total number of cells harvested per vial(T75)by the traditional enzyme digestion method is 4.53×106 cells and the viability rate is 95.5%,while the total number of cells harvested by the enzyme digestion method with polysaccharide of Polygonatum odoratum is 5.35×107 cells and the viability rate is 97.9%;there is no significant difference between hUC-MSCs obtained by the two methods of directional induction differentiation.Conclusion:Compared with the traditional enzyme digestion method,the enzyme digestion method with polysaccharide of Polygonatum odoratum can increase the cell yield and shorten the production time,which is conducive to the large-scale production of cells,and there is no significant difference in cell differentiation,which could meet the production and clinical needs.
