高效液相色谱法测定林芝市藏丹参丹参酮ⅡA的含量

目的:运用高效液相色谱法测定西藏林芝市 4 个藏丹参资源丰富区内藏丹参的丹参酮ⅡA的含量.方法:色谱条件为Supersil ODS2-C18 色谱柱(4.6 mm×250 mm,5 μm),以乙腈-0.02%磷酸溶液为流动相,梯度洗脱,流速 1 mL/min,20℃恒温柱温,检测波长为 270 nm,进样量 10 μL.分别采用超声提取和醇提水沉法提取藏丹参中的丹参酮ⅡA,比较两种方法及不同产地之间的含量差异.结果:超声提取和醇提水沉两种方法均可提取到丹参酮ⅡA,醇提水沉提取量更高,更加适合丹参酮ⅡA的工业化生产.对四个优势产区进行含量检测,米林市的样品含量相对稳定,朗县的样品含量更高.结论:本方法简单准确、高效、稳定性好,为藏丹参的资源开发和种植提供了参考,并为藏丹参资源的充分利用提供了科学依据.
Objective:Determination of tanshinone ⅡA in Salvia castanea Diels f.tomentosa Stib.in four resource-rich areas of Linzhi city by high performance liquid chromatography.Methods:The chromatographic conditions are as follows:Supersil ODS2-C18 column(4.6 mm×250 mm,5 μm),acetonitrile-0.02%phosphoric acid solution as gradient eluent mobile phase,flow rate at 1 mL/min,20℃constant column temperature,detection wavelength of 270 nm,injection volume of 10 μL.Tanshinone ⅡA in Salvia castanea Diels f.tomentosa Stib.is extracted by ultrasonic extraction and alcohol extraction and water sedimentation method,respectively,and the content differences between the two methods and different origins are compared.Results:The experimental results show that the two methods of ultrasonic e xtraction and alcoholic precipitation could extract tanshinone ⅡA,and the content of alcohol extraction and water precipitation is higher,which is more suitable for the industrialized production of tanshinone ⅡA.The content of the four advantageous production areas is tested,in which,the sample content of Milin city is relatively stable,and the sample content in Lang county was even higher.Conclusion:This method is simple,accurate,efficient and stable,which provides a reference for the resource development and cultivation of Salvia castanea Diels f.tomentosa Stib.and a scientific basis for the full utilization of Salvia castanea Diels f.tomentosa Stib.resources.
