Chemical Constituents from Flowers of Polygala fallax and Their Cholesterol-Lowering Activity in Vitro
This work is to investigate the chemical constituents and their cholesterol-lowering activity of flowers of Polygala fallax.Eleven compounds are isolated and purified through silica gel,MCI,Sephadex LH-20 and high-performance liquid chromatography(HPLC)from the low polarity components of 95%ethanol extract of flowers of Polygala fallax.Based on the spectral data and literature comparison,their chemical structures are identified as 3,4-dimethoxy cinnamic acid(1),1,3,6-trihydroxy-2,7-dimethoxyxanthone(2),7-hydroxy-1-methoxy-2,3-methylenedioxyxanthone(3),1,7-dihydroxy-2,3-methylenedioxyxanthone(4),1-methoxy-2,3-methylenedioxyxanthone(5),1,3-dihydroxy-2-methoxyxanthone(6),(2S,3S,4R,10E)-2-[(2R)-2-hydroxyl-tetracosanoly-amino]-10-octadecene-1,3,4-triol(7),aralia cerebroside(8),3-O-β-D-glucopyranosyl spinasterol(9),palmitic acid(10),stearic acid(11),respectively.Compounds 7 and 9 are isolated from this plant for the first time.The effect of the obtained compounds on the cholesterol content are determined using the kit method in HepG2 cells,and the activity screening results show that compound 8 has good cholesterol-lowering activity with an half maximal inhibitory concentration(IC50)of 40.3 μmol/L,which can be used as a candidate molecule for the development of cholesterol-lowering drugs.
