Dscam蛋白在果蝇天然免疫中的作用机制研究

果蝇(Drosophila melanogaster)借助其天然免疫反应来对抗外源病原菌的侵袭.Dscam基因编码的蛋白质是一种细胞粘附分子,最先在神经系统的研究中被发现.最近的研究表明,Dscam基因在免疫系统中也扮演着重要的角色,参与了免疫反应中对病原体的识别和抵抗.本研究利用UAS/GAL4 系统和RNAi技术获得Dscam基因敲低的突变体,以大肠杆菌、藤黄微球菌感染Dscam基因敲低的突变果蝇品系,分析突变体果蝇Dscam基因和抗菌肽基因相对表达量.结果显示,Dscam基因敲低会显著抑制Toll信号通路的表达.
Drosophila melanogaster utilize their natural immune response to combat invasion by exogenous pathogens.The protein encoded by the Dscam gene is a cell adhesion molecule,which was first discovered in research on the nervous system.Recent studies have indicated that the Dscam gene also plays an important role in the immune system,participating in the recognition and resistance against pathogens during immune responses.In this study,Dscam gene knockdown mutants are obtained using the UAS/GAL4 system and RNAi technology,Dscam gene knockdown mutants are infected with Escherichia coli and Micrococcus luteus to analyze the relative expression levels of the Dscam and antimicrobial peptides gene.The results show that Dscam gene knockdown significantly inhibited the expression of the Toll signaling pathway.
