摘要

目的:考察桃红四物汤颗粒的制剂处方及工艺参数.方法:对无辅料干膏粉的性质进行考察,判断颗粒剂制备的可行性.以乳糖、玉米淀粉、糊精和麦芽糊精为辅料,以制粒情况、一次成型率、颗粒性状、颗粒溶化性为考察指标进行辅料种类和用量的筛选.结果:采用无辅料干膏粉直接制粒,制备得到的颗粒可压性一般,流动性良好,有引湿性,可用于干法制粒.优选的辅料为麦芽糊精,载药量在 50%~90%,制备得到的颗粒大小均匀,流动性均较好;颗粒的临界相对湿度为 66%~72%,实际生产中应将外部环境的相对湿度控制在 65%以下.3 批工艺验证结果表明,采用优选的制粒方法制备得到的颗粒收率较高.结论:优选的桃红四物汤颗粒制备工艺稳定可靠,可用于后续桃红四物汤颗粒产业化生产中.

Abstract

Objective:The best drug load and process parameters of Taohong Siwu Tang granules are investigated.Methods:Firstly,the properties of the dry paste powder are investigated to determine the feasibility of the preparation of granules.Lactose,microcrystalline cellulose,corn starch,dextrin and maltodextrin are mixed with dry paste powder respectively,and the types and content of excipients are screened by granulation condition,primary molding rate,particle character,solubility,etc.Results:The dry paste powder is used for direct granulation,and the prepared particles are generally compressible,with good fluidity and hygroscopicity,which can be used for dry granulation.The preferred excipient is maltodextrin,the drug loading is in the range of 50%to 90%,the prepared particles are uniform in size and good fluidity;the critical relative humidity of the particles is 66%to 72%,and the relative humidity of the environment should be controlled below 65%in actual production.The results of the three batches of process validation showed that the particle yield is higher by the preferred granulation method.Conclusion:The preferred preparation process of Taohong Siwu Tang granules are stable and reliable,and can be used in the subsequent industrial production of peach soup particles.
