摘要

以金花茶茶花为原料,采用超声波辅助酶法提取金花茶总黄酮,通过单因素试验和正交试验确定最佳提取工艺,并研究金花茶总黄酮的体外抗氧化活性.结果表明,超声波辅助酶法提取金花茶总黄酮的最佳工艺为超声时间 65 min、乙醇体积分数 70%、酶解时间 70 min、酶解温度 60℃,该条件下金花茶总黄酮的提取率为 6.36%.体外抗氧化性研究表明,总黄酮的抗氧化活性随着质量浓度的增加而增强.金花茶总黄酮含量为 0.20 mg/mL时,对DPPH自由基的清除率为 70.1%,对ABTS自由基的清除率为 91.6%,表现出良好的抗氧化活性.表明超声波辅助酶法提取金花茶总黄酮效果较好,提取效率较高,可为金花茶的进一步研究提供参考.

Abstract

The total flavonoids of Camellia petelotii are extracted by ultrasonic-assisted enzymatic method using Camellia petelotii as raw material,and the optimal extraction process is determined by single factor test and orthogonal test,and the antioxidant activity of total flavonoids of Camellia petelotii in vitro is studied.The results show that the optimal process for extracting total flavonoids from Camellia petelotii by ultrasonic-assisted enzymatic method is 65 min for ultrasonic time,volume fraction of ethanol is 70%,70 min for enzymatic hydrolysis time and 60℃for enzymatic hydrolysis temperature,and the extraction rate of total flavonoids from Camellia petelotii is 6.36%.In vitro antioxidant studies have shown that the antioxidant activity of total flavonoids increases with increasing mass concentration.When the total flavonoid content of Camellia petelotii is 0.20 mg/mL,the scavenging rate of DPPH·radical is 70.1%and the scavenging rate of ABTS·radical is 91.6%,showing good antioxidant activity.The results show that the ultrasound-assisted enzymatic method have a good effect and high extraction efficiency of total flavonoids from Camellia petelotii,which can provide a reference for further research on Camellia petelotii.
