摘要
为了探究菊苣内生真菌的多样性及群落结构,本试验采用高通量测序技术对菊苣叶、根、茎中内生真菌进行真菌的ITS序列测序.结果显示,样品中真菌多样性测序共得到 221 个OUT序列,分属 7 门、23 纲、43 目、72 科、111 属,其中未分类的真菌门、担子菌门和子囊菌门的真菌是门水平上的优势菌门.未分类的真菌属(unclassified_k__Fungi)、链格孢属(Alternaria)在菊苣叶和茎样品中的检测率 90%以上,作为菊苣茎和叶样品中的优势属;unclassified_k__Fungi和纹枯菌属(Rhizoctonia)、Alternaria、刺盘孢属(Colletotrichum)、禾状囊壳属(Poaceascoma)分别在3份根样品中检出率比较高,作为根样品中的优势属.Alpha多样性分析显示,菊苣根样品内生真菌群落的丰度最高,其次苣叶,菊苣茎样品的丰富度最低;而菊苣根样品多样性最丰富,其次是菊苣茎样品,菊苣叶样品多样性最低.主坐标分析结果表明菊苣根、茎、叶样品内生真菌菌落组成上差异比较大.丰度前 20 属的群落Heatmap图显示根样品内真菌多样性以及丰度均较高,而大部分茎和叶样品内生真菌群落相似性高.本研究分析了菊苣不同组织内生真菌多样性和群落结构,为菊苣内生真菌资源的开发利用及其他研究提供了理论依据.
Abstract
To investigate the diversity and community structure of endophytic fungi in Cichorii herba,this study use high-throughput sequencing technology to analyze the ITS sequences of endophytic fungi in leave,root and stem tissue samples of Cichorii herba.Sequencing data suggests that the total of 221 OTU sequences obtain from the all tissue samples,which are classify as 7 classes,23 orders,43 genera,72 family and 111 species.Among them,unclassify fungi,basidiomycetes and ascomycetes are the dominant phyla at the phyla level.unclassified_k__Fungi and Alternaria are detect in more than 90%of the leaf and stem samples,serving as the dominant genera in the leaf and stem samples of Cichorii herba;unclassified_k__Fungi and Rhizoctonia,Alternaria,Colletotrichum,Poaceascoma are the highest detection rates in the three root samples,respectively,serving as the dominant genera in the root samples of Cichorii herba.Alpha diversity analysis show that the root sample of Cichorii herba has the highest diversity of endophytic fungi communities,follow by the leaf sample,and the lowest diversity is found in the stem sample.The root sample is the most diverse,follow by the stem sample,and the leaf sample is the least diverse.The principal coordinates analysis results show that there are significant differences in the composition of endophytic fungi communities in the root,stem,and leaf samples.The heatmap of the top 20 abundant species show that the root sample has the highest diversity and abundance of fungi,while most of the stem and leaf samples have similar endophytic fungi communities.This study analysis the diversity and the community structure of endophytic fungi in different tissues of Cichorii herba,providing theoretical basis for the development and utilization of endophytic fungi resources of Cichorii herba and other studies.
