摘要

电荷异质性作为治疗性蛋白的关键质量属性之一,对药物的安全性和有效性具有重要影响.合理的检测方法可以有效支持上下游工艺开发以及保证治疗性蛋白生产过程中的质量.近年来,对电荷异质性的分析重点逐渐从分析电荷异质体的相对定量发展为对其进行结构表征和活性测定.本文对电荷异质性的来源和结构、经典分析方法以及生物活性分析方法进行了综述,详细阐述了离子交换色谱(IEX)、全柱成像等电聚焦毛细管电泳(WCID-CIEF)在结构表征上的研究进展,旨在为电荷异质性检测提供方法参考.

Abstract

Charge heterogeneities are crucial quality attributes of therapeutic proteins and have profound influence on the safety and efficacy.Selecting the reliable detection methods are essential to support both upstream and downstream process development and manufacturing of therapeutic proteins.In recent years,the focus of charge heterogeneities analysis has gradually developed from the analysis of charge heteromers to structures,and biological activity assessments.In this paper,we review the the source,structural characterization,classical analytical methods and activity analysis analysis methods of charge heterogeneities and recent advancements in ion exchange chromatography(IEX),whole column imaging detection capillary isoelectric focusing(WCID-CIEF)for the qualitative analysis of charge variants,aiming to provide methodological references for drug development.
