As an ideal therapeutic and diagnostic molecule, antibodies are one of the best-selling drugs and have quite good efficacy in the treatment of diseases, especially cancer. As a traditional method of antibody development, the limitations of hybridoma technology limit the progress in the field of antibodies. With the development of molecular biology, simple and effective antibody development methods, such as phage display and yeast surface display, have become an ideal alternative to traditional methods to generate, screen, and isolate high-affinity antibodies with low immunogenicity by using different combinatorial antibody libraries and different selection strategies, which has greatly facilitated the development process of antibody drugs. In this paper, phage and yeast surface display-derived antibodies for cancer, and the differences of phage and yeast display are reviewed to provide reference for research in other disease treatment areas and the development of novel antibodies,as well as provide guidance for the selection and application of the two technologies.
