Study on Synergistic Effect of Zinc Compounds in Anti-dandruff Shampoo Containing Piroctone Olamine
Using piroctone olamine(OCT)as the antibacterial and oil-controlling component,the synergic antibacterial and oil-controlling effects of different zinc compounds on OCT in the traditional sulfate-containing silicone oil system are investigated by using the antibacterial zone of Malassezia furfur,molecular dock prediction and NADPH fluorescence determination methods.The results show that the hair products containing 0.5%OCT has high sensitivity to Malassezia furfur,but zinc compounds have no synergistic effect on OCT,and zinc carbonate,zinc laurate and zinc ricinoleate have antagonistic effect on OCT.Pentanediol,hexanediol and decanediol also have no positive synergistic effect on OCT and zinc gluconate compositions.Lactose zinc,PCA zinc and zinc phenolsulfonate have good results in Docking score index,and zinc ricinoleate and zinc laurate have good results in MMGBSA dG Bind index,indicating more stable binding with 5α-reductase I.Two zinc compounds,0.8%zinc ricinoleate(p<0.05)and 0.8%yeast zinc(p<0.001),show significant synergistic effect on OCT in the fluorescence test of hydrogen donor reduced coenzyme Ⅱ,and have a certain correlation with molecular docking prediction results.The results show that the synergistic compounding mechanism of zinc compounds with OCT is mainly through enhancing the inhibition of 5α-reductase,rather than enhancing the antibacterial effect on Malassezia furfur.Both inorganic zinc and organic zinc have significant effects,especially zinc ricinoleate and yeast zinc.
