Preparation and Performance of γ-Polyglutamic Acid Dressing
Objective:To prepare a multifunctional dressing can be used to heal post-laser surgery wounds.Methods:Hydrogen peroxide-induced L929 cells are used as the oxidative damage model,and the antioxidant damage ability of γ-polyglutamic acid at different molecular weights and concentrations is evaluated by MTT method,and the moisturizing properties,safety and wound healing ability of γ-polyglutamic acid dressing are evaluated by skin occlusion patch test and repetitive open application test.Results:The cell survival rate is 59.73%±8.71%when the concentration of H2O2 is 0.8 mmol/L,and the damage oxidation model of L929 cells is established at this concentration.The cell survival rate is significantly improved when γ-PGA is stimulated by H2O2 at 0.5~2.0 mg/mL,and the comparative study of γ-PGA with different molecular weights shows that H-γ-PGA has the best antioxidant effect in repairing cell damage.Gamma-pga dressings A,B and C are prepared with different concentrations of 1.0,1.5 and 2.0 mg/mL H-γ-PGA respectively,human closed patch test and repeated open smear test shows that subjects has negative skin reactions to γ-PGA dressings A,B and C,and no adverse reactions are observed;the changes of water content in stratum corneum of γ-PGA dressings A,B and C within 8 h are higher than 15%,and the change rates of transepidermal water loss are lower than 15%,indicating that γ-PGA dressings A,B and C has good moisturizing property and the ability of skin barrier repair;the wound healing of γ-PGA dressing(duration of redness and swelling,time of scab shedding)is better than that of normal saline group and blank control group,with statistical significance.Conclusion:The γ-polyglutamic acid dressing prepared in this study exhibits outstanding moisturizing,antioxidant,skin barrier repair,excellent human safety,and wound healing capabilities,and has the potential to be used in wounds after laser surgery.
