Parameter Optimization of AB-PAS Staining Method in Long Toxicity Exploration Experiment of SD Rats
Objective:To optimize the method of AB-PAS staining in frozen sections of gastric and intestinal tissue in long toxicity exploration experiment of SD rats.Methods:The parameters such as the type and fixation time,AB staining time,and experimental water type in the conventional AB-PAS staining step are screened and optimized.Results:The main improved method is set as fixation with Carnoy solution for 1 min,experimental water as ultra-pure water,AB staining time as 5 min,and other conditions remained unchanged.This optimization method could ensure that the histological cell shape are good,the operation repeatability is stable,and the staining results of the sections are clear,the background is clean,and the colors are more vivid.Conclusion:This combination of parameters can be used for the identification of mucous proteins in stomach,intestine and other tissues in SD rat long toxicity exploration experiment,assist in evaluating the degree of tissue lesion.
