Study on the Process of 20nm Membrane Filtration of Human Coagulation Factor Ⅸ
Objective:To optimize the filtration conditions of human coagulation factor Ⅸ nanomembrane and determine the optimal filtration process.Methods:Planova 20N is used to filter human coagulation factor Ⅸ,and the effects of ionic strength,albumin content,arginine content and temperature on the recovery and filtration capacity of human coagulation factor Ⅸ are studied.Results:Under the conditions of albumin content of 2 g/L,arginine content of 5 g/L,ionic strength of 0.3 mol/L,and temperature of 20℃,the 20 nm membrane has the best effect on filtering human coagulation factor Ⅸ.Under this condition,the total filtration amount of 4 h reach 158.58 L/m2,the average filtration rate is about 39.645 L/(m2·h),and the average titer recovery rate of human coagulation factor Ⅸ is 94.98%.Conclusion:By optimizing the filtration conditions,the filtration capacity and recovery rate can be higher,which is conducive to reducing the production cost.
