Determination the Content of Rutin and Astragaline in Different Parts of Potentilla discolor Bge.by HPLC
Objective:To establish an efficient method for determine the content of rutin and astragaline in different parts of Potentilla discolor Bge..Methods:Inertsustain® C18(150 mm×4.6 mm,5 μm)column,methanol-0.3%phosphoric acid aqueous solution as mobile phase,gradient elution,injection volume of 10 μL,detection wavelength of 349 nm,flow rate of 1.0 mL/min,column temperature of 30℃,the contents of rutin and porphyrin in the whole grass,leaves and roots of Potentilla discolor Bge.are determined.Results:The linear relationship is good in the range of 83.8 to 753.8 μg of rutin(R2=0.999 7),and the average recovery rate is 97.57%;the linear relationship is good in the range of 82.9 to 746.2 μg of astragaline(R2=0.999 2),and the average recovery rate is 98.16%.The contents of rutin in whole grass,leaves and roots are 0.710 5 mg/g,0.477 9 mg/g and 0.071 7 mg/g,respectively;the contents of astragaline are 0.621 3 mg/g,0.283 7 mg/g and 0.065 3 mg/g.Conclusion:The rutin and astragaline are mainly concentrated in the leaf parts;the established method is simple,rapid,with good separation effect and stability,and is suitable for the determination of rutin and astragaline content in the Potentilla discolor Bge..
