Studies on spore germination conditions and process of Paenibacillus polymyxa ZJ-9
Paenibacillus polymyxais is widely used in agriculture due to its excellent ability for disease-resistance and growth promotion.The special stress resistance of spores makes them convenient for transportation and storage,however the low germination rate of P.polymyxa under natural conditions greatly limits its general application.Herein,we evaluated the thermal activation conditions and investigated the corresponding germination process of P.polymyxa ZJ-9 spores.As a result,the survival rate of spores reached over 98%under the thermal activation conditions of 75 ℃ for 30 min.More importantly,the subsequent treatment in a mixed solution of L-alanine and sucrose(10.0 mmol/L)in phosphate buffered saline(PBS)led to the germination rate of 63.9%after 12 hours at room temperature.Based on our investigations of the germination process,the addition of L-alanine increased the release of calcium 2,6-pyridinedicarboxylate(DPA-Ca2+)from spores by 3.35 times.During this process,the enhance drehydration of spores was observed by transmission electron microscopy(TEM),and L-alanine greatly promoted the spore germination.This study will be beneficial for further application of P.polymyxa spore.
