SH-SY5Y Cell Damage Caused by Lycium Barbarum Polysaccharide Alleviating H2O2 by Regulating Mitochondrial Function
Objective To explore the mechanism of SH-SY5Y cell injury caused by lycium barbarum polysaccharide(LBP)al-leviating hydrogen peroxide(H2O2)based on mitochondrial damage.Method Human neuroblastoma SH-SY5Y cells were divided into three groups:PBS control group,H2O2 group,and H2O2+LBP group.MTT assay was used to detect cell activity;TUNEL stain-ing was used to detect cell apoptosis;ATP detection kit was used to evaluate ATP levels;Immunofluorescence staining was used to detect the expression of caspase3;Western blot was used to detect the expression of p-Drp1,OPA1,NDUFB8,SDHB,MTCO1,ATP5A,and UQCRC2.Result Compared with the control group,the cell activity of H2O2 group decreased;the apoptosis rate in-creased,the ATP level decreased,the expression of caspase3 and p-Drp1 increased,the expression of OPA1,NDUFB8,SDHB,MT-CO1,ATP5A and UQCRC2 decreased;Compared with the H2O2 group,after LBP intervention,the above indicators showed an oppo-site trend.Conclusion LBP can regulate mitochondrial dynamics,improve mitochondrial function,and alleviate neuronal damage caused by H2O2.
