多指标正交实验优选红芪总黄酮提取工艺的研究

运用正交设计法,以总黄酮的得率及其抗氧化能力为评价指标,考察提取次数、溶剂浓度、提取时间、料液比4个因素对红芪总黄酮提取工艺的影响;紫外吸收法测定红芪总黄酮的含量,DPPH法测定总黄酮的抗氧化能力.结果表明,红芪总黄酮的最佳提取工艺为:药材加8倍量的65%乙醇,回流提取3次,每次40 min.在此条件下红芪总黄酮的提取率25.04%,抗氧化能力最佳,DPPH自由基清除率53.55%.
Multi index orthogonal experiment to optimize the extraction process of total flavonoids of Radix Hedysari
Using orthogonal design method,with total flavonoids yield and the antioxidant capacity of strength as the evaluation index.The effects of extraction times,solvent concentration,extraction time,solid to liquid than the four factors of total flavonoids of Radix Hedysari extraction effect;ultraviolet absorption method for the determination of the content of total flavonoids of Radix Hedysari,DPPH method determination of total flavonoids antioxidant capacity.Results showed that the optimum extraction process of total flavonoids of Radix Hedysari:herbs plus 8 times the amount of 65% ethanol,extracting 3 times,each time 40 min.Under these conditions,the extraction yield of total flavonoids of Radix Hedysari was 25.04%,and the antioxidant capacity is the best,and the DPPH free radical clearance was 53.55%.
