采用离子交联法制备壳聚糖/β-甘油磷酸钠温度敏感性水凝胶,通过正交实验筛选出最优处方.通过研究氯化钠浓度对凝胶时间的影响、凝胶稳定性及降解性实验评价温敏性水凝胶的性能.结果表明,温度敏感性水凝胶最佳制备工艺为:壳聚糖浓度2%、β-甘油磷酸钠浓度40%、HCl浓度0.1 mol/mL、壳聚糖溶液与β-甘油磷酸钠溶液体积比5:1.随着加入的氯化钠浓度增加会延长凝胶时间;该水凝胶在37℃保存72 h后发生变质现象,在-20℃下可长期保存.降解性实验表明其具有良好的生物相容性,具有潜在的应用价值.
Preparation of injectable temperature-sensitive hydrogel and in vitro evaluation
To prepare an injectable chitosan/β-sodium glycerophosphate temperature-sensitive hydrogel by ionic gelation method.The formulation was optimized by orthogonal experiment with gel time as indexes.The properties of temperature-sensitive hydrogel were studied by investigating the experiments of the effect of gel time of hydrogel with different concentration of NaCl and hydrogel' s stabilities and degradation.The results suggested 2% chitosan concentration,β-sodium glycerophosphate concentration 40%,0.1 mol/mL HCl concentration,and the volume ratio of chitosan solution to sodium glycerophosphate solution of 5∶1 was chosen as the optimal formulation to prepare the hydrogel.With increasing of NaCl concentration solution added into hydrogel solution,the gel time was increased.The hydrogel was placed in 37 ℃ condition with 72 h became deteriorated,while the hydrogel still stable in-20 ℃.The degradation experiment of hydrogel indicated good compatible properties and had substantial application.
