Establishment of Efficient and Rapid Regeneration System for Single-flowered Kalanchoe blossfeldiana'Samuba'
The leaves of single-flowered Kalanchoe blossfeldiana were chosen as explants,and orthogonal ex-periments were conducted to investigate the effects of various proportions of different plant growth hormones on the callus induction of leaves,adventitious buds differentiation of callus,root regeneration of seedlings.The transplanting of seedlings was also explored in this study.The results indicated that the optimal medium of K.blossfeldiana callus induction was MS +6-BA 1.0 mg/L +NAA 0.2 mg/L +2,4-D 0.2 mg/L,and the callus in-duction rate was 83.33%.The optimal medium of adventitious bud differentiation from callus was determined to be MS +6-BA 2.0 mg/L + NAA 0.3 mg/L,and the propagation coefficient was 21.4.For the optimal root re-generation of adventitious buds,the medium was 1/2MS +0.2 mg/L +0.2 mg/L,with a rooting coefficient of 7.03.The survival rate of plantlets reached 100%in the meteorites:nutrition soil(1:1)matrix.
