Screening of Saline-alkali Tolerant Bacteria T11 in Potato Rhizosphere and Their Growth Promotion Effect
In order to screen the saline-tolerant bacteria in the potato root zone and explore their growth-promoting effects,bacteria were used as subjects and isolated by the plate streak method.Different NaCl concen-trations(100 g/L,120 g/L,140 g/L,160 g/L,180 g/L)and pH gradients(9,10,11,12,13,14)were set to screen saline-tolerant bacteria in the potato rhizosphere.The 27F/1492-R primers were used to identify the species of bacteria,and their growth-promoting effects were analyzed by measuring the growth curve,the effect of removing salt and alkali,and the ability to produce IAA and protease.The results showed that with the increase of pH value,the tolerance of the bacteria varied.When pH was 13 and the NaCl concentration was 160 g/L,the T11 bacteria survived,while the other bacteria did not grow.The results of the growth promotion assays showed that T11 had the ability to produce IAA and protease,but the effect of removing salt and alkali was weak.The T11 strain belongs to the genus Bacillus and has a growth-promoting effect,laying a foundation for the development of a specialized bacterial fertilizer for potatoes.
