Significance of the expression of prognostic related indicators of renal cancer in nude mice model
Objective Based on the VHL-HIF-EPO signaling pathway,we screened the possible prognostic factors through the intervention of significance on nude mice with clear cell renal cell carcinoma.Methods In this study,786-O cells were injected into nude mice subcutaneously into the back of nude miceto establish rat model.There were 3 groups which were set up according to the period of different drug delivery and one control group.The expressions of Ki67,PCNA and P53 in tumor tissues were detected by HE staining and immunohistochemistry for four times on the 5th,20th and 35th day after intervention and/or natural death,respectively.The expression of related indexes,tumor volume and Fuhrman pathological grade were measured dynamically and survival time was recorded.Results The results of the fourth test between the groups were compared by ANOVE,there were the statistically different in the positive expression rate of Ki67(F=25.91,P<0.001).In the early administration groups,the expression rate of Ki67 was the lowest.Meantime,by Fisher's test,there were the statistically different in the positive expression of PDGFR(x2=12.47,P=0.004)and the mouse mortality(x2=10.34,P=0.006)among different groups.The positive expression of PDGFR and the mouse mortality in the early and middle administration groups was the lowest.Conclusion Early targeted drug intervention can reduce the positive expression of Ki67 and PDGFR,and delay the tumor growth.Therefore,intervention with early targeted drugs may improve the prognosis of RCC patients,and early intervention is necessary in practical work.
