Effect of histone deacetylase inhibitor Tubastatin A on acute liver injury in rats
Objective To investigate the effect of histone deacetylase inhibitor Tubastatin A on acute liver injury in rats.Methods The experimental rats were divided into three groups:normal control group,model group,and intervention group,with 15 rats in each group.The model group and intervention group were induced with D-galactosamine(D-GaIN)/lipopolysaccharide(LPS)to create an acute liver injury model in SD rats.At the same time,the model group was given physiological saline by gavage,and the intervention group was given Tubastatin A.After one week of the experiment,HE staining was used to observe the morphological changes in the rat liver,and the levels of serum glutamyl transferase(GT),aspartate aminotransferase(ALT),alanine aminotransferase(AST),C-reactive protein(CRP),and other indicators were detected.Western blot was used to detect the expression levels of Caspase3 and Bcl-2 in liver tissue.Results Compared with the control group,the model group rats showed significant morphological changes such as edema,necrosis,and inflammatory response in the liver.Tubastatin A intervention could alleviate the degree of liver injury.Compared with the control group,the serum ALT,AST,γ-GT,CRP,IL-6,and other indicators in the model group rats were significantly increased(P<0.05).Tubastatin A intervention could downregulate the levels of serum ALT,AST,γ-GT,CRP,IL-6,and other indicators in rats,and the difference was statistically significant(P<0.05).The expression level of Caspase 3 in the liver of the model group rats was significantly increased,while the expression level of Bcl-2 was significantly decreased.Compared with the model group,the intervention group showed a decrease in Caspase 3 expression level and an increase in Bcl-2 expression level,with statistical significance(P<0.05).Conclusion Tubastatin A has a certain therapeutic effect on acute liver injury induced by D-GaIN/LPS in rats.
