Study on apoptosis induced by Shashensiwei decoction in human lung cancer NCI-H460 cells
Objective:To investigate the apoptosis-inducing effects of Shashensiwei decoction on human lung cancer NCI-H460 cells and to explore the underlying mechanism of apoptosis.Methods:The expression of GADD45a in lung cancer NCI-H460 cells treated with different concentrations of Shashensiwei decoction was detec-ted using immunohistochemistry.The range and quantity of GADD45a in the cells were measured.Flow cytometry(FCM)was used to assess changes in cell apoptosis.Fluorescent quantitative PCR was employed to detect changes in the expression of Caspase-3 and Caspase-7 genes.Results:The expression of GADD45a was significantly increased in the Shashensiwei decoction group,with statistical differences observed between the Shashensiwei decoction groups of different concentrations and the control group.FCM analysis of Annexin-V/PI double staining results showed that the apoptosis rate of cells in the Shashensiwei decoction group was higher compared to the control group(P<0.05).Fluorescent quantitative PCR revealed that the expression of Caspase-3 and Caspase-7 genes in NCI-H460 cells was decreased in the Shashensiwei decoction group compared to the control group(P<0.05).Conclusion:Shashens-iwei decoction exhibits potential in vitro inhibitory effects on the proliferation of human lung cancer NCI-H460 cells.It induces the production of GADD45a,Caspase-3,and Caspase-7 in lung cancer cells,promoting apoptosis in NCI-H460 cells.Therefore,it is inferred that Shashensiwei decoction can inhibit the growth of lung cancer H460 cells.
