Effect of eliminating dampness therapy on gland function and gland pathological state of mice with spontaneous Sj?gren's syndrome
Objective:To approach the mechanism of eliminating dampness therapy on Sjögren's syndrome.Methods:NO D/Ltj mice of spontaneous Sjögren's syndrome model were randomly divided into eliminating dampness group,nourishing yin to eliminate dampness group and model group.ICR mice were selected as control group.The control group and model group were given normal saline by gavage,while the other groups were given corresponding drugs for treatment.The treatment with drug lasted for 16 weeks.The salivary flow rate of mice was detected during the gavage.After the gavage,the mice were sacrificed,the submandibular glands were taken out and pathologically evaluated,and the proportion of marrow-derived suppressor cells(MDSC)of cervical lymph nodes in the spleen and peripheral blood was detected by flow cytometry.Results:Compared with the control group,the salivary flow rate of the mice in model group was significantly decreased(P<0.05 or P<0.01);the proportion of MDSC in peripheral blood was higher than that in the control group(P<0.05);the level of MDSC in the spleen showed a rising trend compared with the control group(P>0.05);and the level of lymphocyte infiltration area to total area in submandibular gland was significantly higher than that in the control group(P<0.05).Compared with the model group,the saliva flow rate of the mice in eliminate dampness group and nourishing yin to eliminate dampness group was significantly increased at 12 weeks of age,18 weeks of age and 21 weeks of age;both in eliminate dampness group and in nourishing yin to eliminate dampness group,the level of MDSC in peripheral blood and spleen was significantly reduced;and in the two groups,the proportion of lymphocyte infiltration area to total area in submandibular gland of mice showed a downward trend,but there was no significant statistical difference(P>0.05).Conclusion:Eliminating dampness therapy can promote the secretion of salivary glands in mice of Sjögren's syndrome model,reduce the level of MDSC in peripheral blood and spleen,and has a significant therapeutic effect on Sjögren's syndrome.
