Approach to protective effect of modified Quzhi decoction on cartilage and meniscus injury in modeled rats with knee joint osteoarthritis
Objective:To approach the possible mechanism of modified Quzhi decoction on relieving the symptoms of knee joint osteoarthritis.Methods:54 SPF male rats were divided into blank group,model group,low-dose modified Quzhi decoction group,middle-dose modified Quzhi decoction group,high-dose modified Quzhi decoction group and celecoxib group,9 rats in each group.Except for the blank group,other groups were made into rat models of knee osteoarthritis by intra-articular injection of sodium iodoacetate,and the drug intervention was conducted for 6 weeks after modeling.The pathomorphological changes of cartilage and meniscus of knee joint in rats were observed by HE staining.The degree of cartilage degeneration was evaluated by Mankin method.The contents of IL-1 β,TNF-α,IL-17,IL-6 and MMP-1 in rat's serum were detected by ELISA.The expressions of IL-6 and MMP-1 in meniscus tissue were detected by immunohistochemistry.The expression levels of NF-κ B and CIKS(NF-κB activator)mRNA in rat's cartilage were detected by PCR method.Results:Compared with the model group,the structure of rat's cartilage in modified Quzhi decoction group was slightly damaged,the surface of cartilage was undersmooth,the distribution of chondrocytes was uneven,and the matrix staining was reduced;the surface of meniscus was rough,the surfaces of some meniscus were broken and torn,the distribution of nuclei was uneven,the volume of nuclei became larger,the focal appearance increased.The expression levels of IL-1 β,TNF-α,IL-17,IL-6 and MMP-1 in serum of rats in modified Quzhi decoction group were lower than those in model group(P<0.05).The results of immunohistochemistry showed that the expression levels of IL-6 and MMP-1 in meniscus tissue were lower than those in model group(P<0.05).Compared with the model group,the expression levels of NF-κB and CIKSmRNA in cartilage of rats in modified Quzhi decoction group were decreased(P<0.05).Conclusion:Modified Quzhi decoction can alleviate the inflammatory reaction in knee joint of KOA rats,regulate the expression of related molecules in IL-17/NF-κB signal pathway,alleviate the degeneration of cartilage and meniscus of knee joint,and achieve the effect of improving knee joint osteoarthritis.
