Efficacy observation of Bushen Huoxue Jiedu formula and umbilical cord mesenchymal stem cell transplantation on rats with acute liver failure
Objective:To observe the therapeutic efficacy of Bushen Huoxue Jiedu formula and rat's umbilical cord mesenchymal stem cells(rUCMSCs)transplantation on rats with acute liver failure(ALF).Methods:rUCMSCs were extracted by two-step digestion method.ALF rats models were established by D-galactosamine/lipopolysaccharide(D-gal/LPS).After 24 hours,40 surviving model rats were randomly divided into TCM and transplantation group,transplantation control group,TCM control group and blank control group.TCM and transplantation group and transplantation control group were injected with 2x 106 rUCMSCs through tail vein,while remaining two groups were injected with equal volume of aseptic PBS solution through tail vein.In addtion,TCM and transplantation group and TCM control group were given Bushen Huoxue Jiedu decoction(10mL/kg)by intragastric administration,while remaining two groups were given equal volume of normal saline by intragastric administration.Meanwhile,the general conditions of rats in each group were observed daily.On the 7th day after treatment,the 7-day survival rate of rats was calculated in each group,and the levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST)and total bilirubin(TBIL)in serum were detected.Then,the histopathological changes of liver and lung were observed by hematoxylin-eosin(HE)staining.Results:After modeling,the survival state of model rats was declined gradually,and the liver function was significantly abnormal.After treatment,the survival rates in TCM and transplantation group,transplantation control group,TCM control group and blank control group were 70％,50％,40％and 20％respectively.The survival rate of TCM and transplantation group was significantly higher than that of blank control group(P＜0.05).The levels of TBIL,ALT and AST in serum of rats in TCM and transplantation group were decreased significantly,and the differences were statistically significant compared with remaining three groups(P＜0.05).Pathological examination showed that the liver tissue of rats in TCM and transplantation group was significantly repaired,the structure of hepatic lobule was intact,the infiltration of inflammatory cells was significantly reduced;furthermore,no adverse reactions such as pulmonary embolism,pulmonary fibrosis,etc.were observed.Conclusion:The combination of Bushen Huoxue Jiedu formula and rUCMCSs transplantation can significantly alleviate liver's inflammation in ALF rats,improve liver function,promote liver regeneration,and increase the survival rate of ALF rats.And this therapeutic effect is better than that of individual application of anyone of the two.The combinating application of Bushen Huoxue Jiedu formula and rUCMCSs transplantation has a synergistic effect on the treatment of ALF.
