Approach to mechanism of Yiqi Chuwen granule on acute lung injury in mice
Objective:To observe the therapeutic effect of Chinese herbal medicine Yiqi Chuwen granule on acute lung injury(ALI)in mice model and approach its potential mechanism,especially in inhibiting inflammation and cytokine storm.Methods:The experiments of in vitro cytotoxicity and pseudovirus inhibition were carried out by 293T-ACE2 cells and Omicron BA.5 pseudovirus.In in vivo experiment,the acute lung injury models of BALB/c mice induced by lipopolysaccharide(LPS)were divided into four groups i.e.normal group,model group,DXMS group and Yiqi Chuwen group.The therapeutic effect of Yiqi Chuwen granule was evaluated by HE staining,PAS staining,immunofluorescence and ELISA etc.The changes of expression levels of cytokine IL-6,IL-10,IFN-γ and TNF-α were observed.Results:Yiqi Chuwen granule showed obvious antiviral activity in in vitro experiment,and could effectively inhibit the cell invasion of Omicron BA.5 pseudovirus.The results of in vivo experiment showed that Yiqi Chuuwen granule obviously alleviated the pathological injury of lung tissue induced by LPS,reduced the infiltration of inflammatory cytokines and improved wet/dry weight ratio of lung tissue(P＜0.05).In addition,this drug could also regulate the immune response,reduce the expression of pro-inflammatory cytokines(P＜0.05),increase the expression of anti-inflammatory cytokine IL-6(P＜0.05),and alleviate the affection of cytokine storm.Conclusion:Yiqi Chuwen granule has a good therapeutic effect on acute lung injury.Its mechanism may involve in inhibiting virus invasion,regulating inflammatory reaction and cytokine expression.
