目的 探讨复发性流产(RSA)患者绒毛组织中EPHB4下调与绒毛血管生成障碍的关系。方法 收集19例RSA患者(RSA组)和14例早孕要求终止妊娠患者(NC组)的绒毛组织样本,通过RT-qPCR和Western blotting检测EPHB4的表达,通过免疫组化染色标记CD31计算绒毛微血管密度,并分析两者间的相关性。结果 RSA组患者的组织中EPHB4的表达、微血管密度低于NC组(P<0。05)。EPHB4 mRNA与绒毛微血管密度成中度正相关(r=0。4961,P=0。0362)。结论 EPHB4在RSA患者中表达下调,可能通过抑制绒毛血管生成参与RSA的发生发展。
Study on the correlation between EPHB4 and villus angiogenesis in recurrent abortion
Objective To investigate the relationship between downregulation of EPHB4 in villus tissue and dysangiogenesis in patients with recurrent abortion(RSA).Methods Villus tissue samples from 19 RSA patients(RSA group)and 14 early pregnancy termination patients(NC group)were collected.The expression of EPHB4 was detected by RT-qPCR and Western blotting.The villus microvascular density was calculated by immunohistochemical staining labeled CD31,and the correlation between the two samples was analyzed.Results The expression of EPHB4 and microvascular density in RSA group were lower than those in NC group(P<0.05).EPHB4 mRNA was moderately positively correlated with villus microvessel density(r=0.4961,P=0.0362).Conclusion The expression of EPHB4 is downregulated in patients with recurrent spontaneous abortion,which may be involved in the occurrence and development of RSA by inhibiting villus angiogenesis.
