为了优选玉参花饮料(Yushenhua beverage,YSH)的最佳提取工艺,以总多糖、党参炔苷和绿原酸含量的多指标加权综合评分为评价指标,用苯酚硫酸法测定其多糖含量,采用高效液相色谱法测定其党参炔苷和绿原酸含量.在单因素试验基础上,采用Box-Behnken效应面法,对YSH原料的浸泡时间、料液比和提取时间等工艺条件进行优化,得到YSH的最佳提取工艺为:浸泡时间 15 min、料液比 1∶14 g/mL、1∶12 g/mL,提取时间60 min、40 min,此条件下优选的玉参花饮料提取工艺合理可行.
Study on the extraction process of Yushenhua beverage
To optimize the extraction process of Yushenhua beverage(YSH),the multi-indicator weighted scores of total polysaccharides,lobetyolin and chlorogenic acid content were used as the evaluation index.The polysaccharides content was determined by the phenol-sulfuric acid method.The content of lobetyolin and chlorogenic acid was determined by high-performance liquid chromatography method.Based on the one-factor test,the soaking time,material-to-liquid ratio,and extraction time of YSH raw materials were optimized by Box-Behnken response surface methodology.The optimal extraction process of YSH was determined as follows:soaking time of 15 minutes,material-to-liquid ratio of 1∶14 g/mL and 1∶12 g/mL,and extraction time of 60 minutes and 40 minutes.The preferred extraction process of YSH beverage was found to be reasonable and feasible under these conditions.
