Exploration and practice of comprehensive experiment of modern pharmaceutics course design:A case study on resveratrol
[Objective]A comprehensive experiment of modern pharmaceutics is a vital part of the pharmacy course at Sun Yat-sen University,which is"Double First-Class,"because it enhances the professional competence of undergraduate students comprehensively.The course has been practiced for numerous years,and good teaching results have been recorded.To summarize and improve the overall experience,an experiment involving the extraction and separation,quality analysis of resveratrol,preparation and quality analysis of formulations,and teaching discussions was conducted using resveratrol as an example.[Methods]The comprehensive experiment of modern pharmaceutics primarily involves the following stages.First,the responsible teachers designate the compound to be extracted or synthesized,and the students choose the method,team up,and then analyze and summarize the literature and draft an initial scheme.Then,the students revise the experimental scheme under the guidance of the teachers and determine the experimental scheme after verifying its feasibility through preliminary experiments.Next,they enter the experimental phase as follows.First,resveratrol is extracted using either the alkaline extraction-acid precipitation method or the ultrasound-assisted enzymatic hydrolysis method according to the students'experimental scheme.Resveratrol is separated and purified by extraction and silica gel column chromatography,and the structure of the product is analyzed and identified using an infrared(IR)spectrometer and nuclear magnetic resonance spectrometer(NMR).Second,the quality evaluation of the raw materials,including the description,identification,loss on drying,residue after ignition,heavy metal,and content determination,is conducted.The high-performance liquid chromatography method is realized to determine the resveratrol content.Third,resveratrol microemulsion and resveratrol microemulsion gel are prepared according to the prescribed formula and process.The quality analysis of the pharmaceutical preparations is conducted,including the appearance inspection,particle size diameters,stability,pH,viscosity and content determination.Once all steps are implemented,students write course papers and summarize their reports.Finally,a teaching discussion is conducted on the selection criteria of compounds and the experimental teaching arrangements.[Results]The extraction yield of resveratrol using the alkaline extraction-acid precipitation method and the ultrasound-assisted enzymatic hydrolysis method was 0.363% and 0.275%,respectively.The structure of the product was confirmed as resveratrol through the IR and NMR.The quality of resveratrol raw material satisfied the standards in terms of description,identification,loss on dry,residue after ignition,and heavy metals.The resveratrol exhibited a good linearity in the range of 12.5-62.5 μg/mL(R2=0.9998).Further,it possessed good system suitability,specificity,and repeatability.The content of resveratrol prepared by the students was in the range of 53.73%-94.56% .The resveratrol microemulsion gel was a colorless,transparent gel with an average particle size of 8.842 nm.It yielded good stability,pH of 5.0-6.0,and an average viscosity of 27 600 mPa·s.Further,the average content of resveratrol was 95.94% (n=3).[Conclusions]According to the main process of drug development and production,a comprehensive experiment of modern pharmaceutics is composed of several independent experimental courses such as pharmaceutical chemistry,chemistry of natural products,pharmaceutical analysis,and pharmacy.Resveratrol and resveratrol microemulsion gel prepared by the students according to the experimental scheme satisfied the required standards.Through the entire process of participating in experiment design,experiment,post-experiment summary,and discussion,students would become interested in scientific research and enhance their ability to innovate proactively.In addition,by combining practice with theory,students would learn to analyze and solve problems comprehensively.In addition,students would enhance team cooperation spirit and overall planning abilities as well as the spirit of cooperation,laying a solid foundation for future scientific research and work.
