Genetic Diversity Among Morchella conica Based on AFLP Analysis
Forty-nine single ascospore isolates and one tissue isolate obtained from a single fruit body of Morchella conica were separated into four groups based mycelial colony morphology and subjected to genetic diversity analysis using AFLP markers. Polymorphic loci represented by 488 distinct bands ranging in size from 100 to 600 bp were detected using four screened primer pairs, indicating abundant genetic diversity among the isolates. UPGMA-based clustering separated the 50 test isolates into 11 groups. Further analysis of five morphologically distinct groups (SS, NS, SN, NN, MB) and the 11 UPGMA-based groups was undertaken using the 1.32 version of POPGENE software.
