尖顶羊肚菌遗传多样性的AFLP分析

以是否产菌核及产菌斑为标准将尖顶羊肚菌子实体ZD-18分离的49个单孢菌株分为四组,并以1个组织分离菌株为对照,采用AFLP技术进行DNA多态性分析.用筛选出的4对引物进行扩增,在100 bp-600 bp有效带范围内的488条带都具有多态性,通过Nei's无偏遗传相似度及遗传距离(1978)的UPGMA聚类分析,可将50个菌株分为11个组群,且与四个形态分组的多态性指数很一致,但形态和遗传仅在产菌斑性状上具有对应趋势,产菌核性状的遗传聚类说明其基因表达复杂多变,可利用研究得出的多态性遗传图谱进行较深入的分子研究.
Genetic Diversity Among Morchella conica Based on AFLP Analysis
Forty-nine single ascospore isolates and one tissue isolate obtained from a single fruit body of Morchella conica were separated into four groups based mycelial colony morphology and subjected to genetic diversity analysis using AFLP markers. Polymorphic loci represented by 488 distinct bands ranging in size from 100 to 600 bp were detected using four screened primer pairs, indicating abundant genetic diversity among the isolates. UPGMA-based clustering separated the 50 test isolates into 11 groups. Further analysis of five morphologically distinct groups (SS, NS, SN, NN, MB) and the 11 UPGMA-based groups was undertaken using the 1.32 version of POPGENE software.
