Determination of Polarity of Naematelia aurantialba by Single Spore Hybridization
Basidiospores were collected from mature fruiting bodies of Naematelia aurantialba,and eventually a total of 50 yeast-like single spore isolates were obtained through single spore isolation and nuclear staining.Co-cultivation of such a yeast-like single spore isolate with Stereum hirsutum failed to produce fruiting bodies.The first round of mating test showed that the compatibility was 24.5%.After another three rounds of mating tests,it was determined that there were four mating types,A1B1(13),A1B2(13),A2B1(12),and A2B2(12),suggesting that N.aurantialba belongs to quadrupole heterothallism fungi.Then five single spore isolates were randomly selected from each mating type for pairwise mating and cultivation experiments.The results showed that mature fruiting bodies of N.aurantialba were developed when co-culturing single spore isolates that can form dikaryons with the mating type A1A2B1B2 together with S.hirsutum.
